SRX14122053: PacBio CLR whole genome sequencing of Helicoverpa zea: male pupa

Design: DNA extraction was performed from a single male (homogametic, ZZ sex chromosome) whole insect (ilHelZeax1) using the MagAttract HMW DNA Kit modified from manufacturer instructions to include gentle inversion to mix all components, an additional wash step, and eluting DNA from beads with three separate additions of 115.0 l AE buffer at 37C. Un-sheared DNA was prepared for PacBio sequencing using the SMRTBell Express Template kit 2.0. Sequencing was performed on a Sequel II System using Binding Kit v2.0, Sequencing kit v2.0, and SMRT Cell 8M. To target CLR reads, the library was sequenced using a 20 hour movie time on one SMRTcell.
